Furthermore, we detail the influence of substituents on molecular structures, frontier molecular orbital energy levels, and the magnetic ring current tropism they induce. Theoretical calculations and X-ray structural analyses concur that derivatives of the C2h-substitution pattern display diverse C2h structures, the degree of bond length alternation being highly dependent on the substituents' electronic properties. A selective modulation of frontier molecular orbital energy levels is observed, as a result of the nonuniformity in their distribution and the influence of electron-donating substituents. The inversion of the HOMO and HOMO-1 sequences, consistent with theoretical predictions and validated by visible and near-infrared absorption spectra, is observable in the intrinsic s-indacene system. The s-indacene derivatives' 1H NMR chemical shifts, coupled with their NICS values, point to a subdued degree of antiaromaticity. The attractive combination of high thermostability, protease resistance, and robust heterologous expression makes encapsulins a popular choice for bioengineering applications, encompassing medicine, catalysis, and nanotechnology. In biotechnological applications, organisms capable of resisting extremes in physicochemical conditions, such as high temperature and low pH, are highly desirable. No systematic investigation into acid-tolerant encapsulins has been conducted, leaving the influence of pH on encapsulin shells unexplored. The acid-tolerant bacterium Acidipropionibacterium acidipropionici is the source of a newly identified encapsulin nanocage, which is detailed in this report. Our analysis, encompassing transmission electron microscopy, dynamic light scattering, and proteolytic assays, showcases its exceptional tolerance to extreme acidity and protease degradation. We utilize cryo-electron microscopy to structurally characterize the novel nanocage, finding a dynamic five-fold pore displaying both closed and open states at a neutral pH, but only a singular closed state under the effects of strongly acidic conditions. The open state, notably, features the largest pore reported in any encapsulin shell to date. The demonstrated capability of non-native proteins to be encapsulated is examined, along with the study of external pH levels affecting the internal cargo. Annually, approximately 10,000 new cases are documented in Mexico. The IMSS, a pioneer in HIV care, has progressively adopted various antiretroviral therapies for people living with HIV. The 1990s saw the initial implementation of zidovudine at an institutional level as an antiretroviral, with the subsequent integration of other agents, including protease inhibitors, non-nucleoside reverse transcriptase inhibitors, and integrase inhibitors. Antiretroviral therapy schemes, successfully integrated in 2020 into a single-tablet format based on integrase inhibitors, demonstrated exceptional efficacy, delivering treatment to 99% of the population in a timely and efficient manner. From a preventative standpoint, the IMSS has been at the forefront, being the first institution to implement national HIV pre-exposure prophylaxis in 2021 and, subsequently, providing universal post-exposure prophylaxis in 2022. With the intention of improving the lives of people living with HIV, the IMSS remains at the forefront of incorporating different management tools and instruments.
